Quantitative polymerase chain reaction (qPCR) identifies DNA from pathogens commonly associated with mastitis and has various benefits. Rapid identification of mastitis-causing pathogens (approximately 3 hours), including pathogens that cannot be grown using conventional culturing techniques; objective reporting of DNA found in milk samples; and the ability to detect non-living pathogens. The commercial qPCR kit detected Mycoplasma bovis, Mycoplasma spp., Staphylococcus aureus, coagulase-negative staphylococci (CNS), Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis, Prototheca spp., Escherichia coli, Klebsiella spp. The prevalence of specific DNA is determined by its presence or absence in samples, taking into account both sampling protocols and herds. In dairy cows, pathogens in the mammary gland cause intramammary infection, which can progress to clinical or subclinical mastitis. Pathogens that cause mastitis can be classified as contagious, environmental, or opportunistic based on their primary reservoirs and mode of transmission. Infected tissues, such as the udder, serve as the primary substrate for contagious pathogens. qPCR can also be used in milk samples treated with conventional preservatives, such as the bactericidal Bronopol (2-bromo-2-nitropropan-1,3-diol). When compared to bacterial culture, qPCR significantly reduces analysis time and enables the simultaneous testing of a large number of milk samples for a variety of pathogens.
